Sample diletakkan dalam test device dan hasilnya akan muncul dalam waktu 5 &#8211; 30 menit. Teknologi ini dapat digunakan untuk mendeteksi secara cepat kandungan antibodi, antigen, asam nukleat, dan analit lainnya dalam sampel. Lateral Flow Assay Device merupakan perangkat pengujian diagnostik yang mudah digunakan, portable, dapat dilakukan secara mandiri, cepat, dan murah. Perangkat uji rapid test dapat disimpan pada suhu ruang dalam jangka waktu yang lama, tidak memerlukan pemrosesan sampel terlebih dahulu, dan memberikan hasil diagnose tanpa pemrosesan sampel atau alat tambahan, menjadikan teknologi ini sangat idel untuk point-of-care dan field-based diagnostics.<\/p>\n<p class=\"text-justify\">Konfigurasi pengujian LFA yang paling umum menggunakan colloidal gold nanospheres yang dapat menghasilkan sinyal kolorimetrik yang dapat dilihat langsung sinyalnya tanpa menggunakan alat pembacaan khusus. Sinyal kolorimetrik inilah yang muncul dan terlihat pada garis test line dan control line (Gambar 1)<\/p>\n<p>&nbsp;<\/p>\n<p><center><img decoding=\"async\" class=\"x-ray-image\" src=\"https:\/\/www.sciencewerke.com\/images\/Screenshot1.png\" alt=\"Sciencewerke\" \/><\/center><center>Gambar 1. Warna merah muncul dari penyerapan cahaya hijau dan biru oleh nanopartikel emas. Karena sifat optik nanopartikel emas bergantung pada ukuran dan bentuk, sensitivitas pengujian dibatasi oleh intensitas sinyal dari partikel reporter kecil (biasanya berdiameter 40 nm). Sensitivitas deteksi analit sangat penting. Jika sensitifitas device LFA tidak mampu mendeteksi analit pada sampel, maka tentu tidak akan laku terjual di pasaran.<\/p>\n<p class=\"text-justify\">Salah satu tipe nanopartikel yang umum digunakan dalam pembuatan rapid test device adalah nanopartikel emas atau biasa juga disebut koloid emas. Nanopartikel emas memiliki sifat optik yang unik yang menjadikannya sebagai penyerap cahaya yang kuat. Jenis nanopartikel emas yang banyak digunakan berukuran 40 nm yang memiliki peak absorbansi pada panjang gelombang ~520 nm, nanopartikel emas ini menghasilkan warna ruby red pada test line dan control line. Permukaan nanopartikel emas secara natural memiliki kemampuan afinitas terhadap antibodi ataupun protein lainnya, sehingga memudahkan untuk melakukan konjugasi antibodi atau protein dengan nanopartikel emas hanya dengan proses pencampuran atau memanfaatkan affinity ligand. Selain nanopartikel emas dengan ukuran 40 nm, ada beberapa ukuran dan bentuk nanoprtikel lainnya yang digunakan sebagai pewarna dalam pembuatan device lateral flow assay, seperti ukuran 60, 80, dan 100 nm.<\/p>\n<p class=\"text-justify\"><strong>Baca juga :<\/strong><a href=\"\/sciencewerke.indo\/pentingnya-syndromic-testing-untuk-identifikasi-penyakit-pneumonia\">\u00a0Pentingnya syndromic testing (multiplex PCR) untuk identifikasi penyakit pneumonia.<\/a><\/p>\n<p class=\"text-justify\">Salah perusahaan pembuat nanopartikel yang berbasis di USA, NanoComposix berhasil membuat nanopartikel emas yg memiliki kemampuan untuk meningkatkan sensitivitas uji\u00a0<strong>hingga 6 sampai 20 kali lipat\u00a0<\/strong>dibandingkan dengan nanopartikel emas 40 nm, nanopartikel tersebut disebut dengan\u00a0<strong>\u201cGold Nanoshell\u201d<\/strong>. BioReady 150 nm gold nanoshell merupakan nanopartikel reporter plasmonik yang terdiri dari inti silika 120 nm yang dilapisi dengan cangkang emas tipis (12-18 nm) (Gambar 2). Bagian dalam 2 embra mengurangi kerapatan partikel 2 embrane terhadap nanopartikel emas padat, memungkinkan flowrate yang lebih baik melalui 2 embrane lateral flow dibandingkan dengan gold nanosphere 150 nm.<\/p>\n<p>&nbsp;<\/p>\n<p><center><img decoding=\"async\" class=\"x-ray-image\" src=\"https:\/\/www.sciencewerke.com\/images\/Screenshot2.png\" alt=\"Sciencewerke\" \/><\/center><center>Gambar 2. Nilai peak extinction nya 35x lebih besar dibandingkan nanopartikel emas 40 nm yang berbentuk spherical (bulat penuh) (Gambar 3).<\/p>\n<p>&nbsp;<\/p>\n<p><center><img decoding=\"async\" class=\"x-ray-image\" src=\"https:\/\/www.sciencewerke.com\/images\/Screenshot3.png\" alt=\"Sciencewerke\" \/><\/center><center>Gambar 3. (A) Gold nanoshell menyerap cahaya lebih banyak per partikel dibandingkan nanopartikel emas 40 nm dan (B) nanopartikel emas 40 nm (kiri) dan gold nanoshell 150 nm (kanan) dengan konsentrasi yg sama.<\/center>&nbsp;<\/p>\n<p class=\"text-justify\">Karena gold nanoshell memiliki nilai particle extinction cross section yang lebih besar dari nanopartikel emas 40 nm, tidak mebutuhkan banyak partikel terikat untuk bisa menghasilkan sinyal yang dapat terdeteksi pada rapid test. Hal ini berarti, sensitivitas dari device yang menggunakan gold nanoshell sebagai pewarna konjugatnya lebih baik dibandingkan dengan device yang menggunakan nanopartikel emas 40 nm. Karena ini sama-sama nanopartikel emas, proses konjugasinya dengan antibodi atau protein lainnya juga tidak jauh berbeda dengan cara konjugasi pada nanopartikel emas biasa dengan sedikit modifikasi saja. Warna yg terlihat pada test line dan control line pada device yang menggunakan nanopartikel emas 40 nm adalah merah bata atau ruby red, sedangkan pada device yang menggunakan gold nanoshell 150 nm berwarna hijau kehitaman, seperti yang terlihat pada Gambar 4 di bawah ini yang menunjukkan peningkatan sensitifitas hingga 20x pada pengujian hormone dengan rapid test device. Device yang menggunakan gold nanoshell 150 nm mampu mendeteksi target hormone hingga 0.16 UI\/L.<\/p>\n<p>&nbsp;<\/p>\n<p><center><img decoding=\"async\" class=\"x-ray-image\" src=\"https:\/\/www.sciencewerke.com\/images\/Screenshot4.png\" alt=\"Sciencewerke\" \/><\/center><center>Gambar 4.
