Droplet membentuk partisi yang memisahkan molekul DNA sampel. Sampel difraksinasi menjadi 20.000 droplet dan amplifikasi PCR dari molekul DNA cetakan terjadi pada setiap individu droplet. Teknologi ddPCR menggunakan reagen dan alur kerja yang serupa dengan yang digunakan untuk sebagian besar pengujian PCR standar berbasis probe TaqMan. Droplet pada dasarnya memiliki fungsi yang sama dengan tabung atau sumur uji individu pada plate tempat reaksi PCR berlangsung, meskipun dalam format yang jauh lebih kecil. Partisi sampel secara besar-besaran adalah aspek kunci dari teknik ddPCR.<\/p>\n<p>&nbsp;<\/p>\n<p><center><img decoding=\"async\" src=\"https:\/\/www.sciencewerke.com\/images\/news\/droplet-digital-pcr-untuk-pengujian-patogen-pada-air-limbah1.png\" alt=\"Sciencewerke\" \/><img decoding=\"async\" src=\"https:\/\/www.sciencewerke.com\/images\/news\/droplet-digital-pcr-untuk-pengujian-patogen-pada-air-limbah2.png\" alt=\"Sciencewerke\" \/><\/center>&nbsp;<\/p>\n<p class=\"text-justify\">Partisi sampel adalah aspek kunci dari droplet digital PCR. Pada PCR tradisional, satu sampel hanya dilakukan satu pengukuran, tetapi pada Droplet Digital PCR, sampel dipartisi menjadi droplet sebanyak 20.000. Partisi ini memungkinkan pengukuran ribuan peristiwa amplifikasi independen dari satu sampel.<\/p>\n<p>&nbsp;<\/p>\n<p><center><img decoding=\"async\" src=\"https:\/\/www.sciencewerke.com\/images\/news\/droplet-digital-pcr-untuk-pengujian-patogen-pada-air-limbah3.png\" alt=\"Sciencewerke\" \/><\/center>&nbsp;<\/p>\n<p class=\"text-justify\">Teknologi ddPCR menggunakan kombinasi mikrofluida dan bahan kimia surfaktan eksklusif untuk membagi sampel PCR menjadi droplet. Amplifikasi PCR terjadi secara independen pada setiap droplet yang mengandung cetakan DNA dan reagen PCR. Setelah PCR, setiap droplet dianalisis atau dibaca untuk menentukan fraksi droplet PCR-positif pada sampel. Data tersebut kemudian dianalisis menggunakan statistik Poisson untuk menentukan konsentrasi cetakan DNA target dalam sampel.<\/p>\n<p class=\"text-justify\">Sistem ddPCR dapat digunakan untuk:<\/p>\n<p><i class=\"fa fa-chevron-circle-right\"><\/i>\u00a0\u00a0Deteksi target DNA prevalensi rendah\/langka dengan sensitivitas tinggi<br \/>\n<i class=\"fa fa-chevron-circle-right\"><\/i>\u00a0\u00a0Menentukan variasi jumlah salinan DNA (copy number) dengan akurasi tinggi<br \/>\n<i class=\"fa fa-chevron-circle-right\"><\/i>\u00a0\u00a0Mengukur tingkat ekspresi gen dengan presisi tinggi<\/p>\n<p>Pengujian SARS-CoV-2 pada Air Limbah dengan Teknologi Droplet Digital PCR (ddPCR)<\/p>\n<p class=\"text-justify\">Epidemiologi berbasis air limbah secara tidak langsung dapat memantau tingkat infeksi di komunitas berdasarkan jumlah patogen dalam air limbah setempat. Karena individu yang terinfeksi SARS-CoV-2, baik simptomatik dan asimptomatik, melepaskan RNA virus dalam sistem saluran pembuangan, analisis SARS-CoV-2 pada air limbah dari saluran pembuangan dapat memberikan informasi tentang infeksi SARS-CoV-2 dalam komunitas. Namun, keakuratan data bergantung pada kualitas metode pengujian air limbah. Kit tersebut direkomendasikan oleh CDC AS dan menggunakan primer RT-PCR WHO untuk deteksi SARS-CoV-2, memungkinkan deteksi gen N2 dan E pada SARS-CoV-2.
